Abstract |
IntracellularIntracellular Ca2+Ca2+ is involved in control of a large variety of cell functions including apoptosisapoptosis and neuronneuron cell deathneuron cell death . For example, intracellular Ca2+ overloadoverload is critical in neuron cell deathcell death induced by excitotoxicityexcitotoxicity . Thus, single cell monitoringsingle cell monitoring of intracellular Ca2+ concentrationintracellular Ca2+ concentration ([Ca2+]cyt[Ca2+]cyt ) in neuronsneurons concurrently with apoptosis and neuron cell death is widely required.Procedures for culture and preparation of primary culturesprimary cultures of hippocampalhippocampal ratrat neuronsneurons and fluorescence imagingfluorescence imaging of cytosolic Ca2+Ca2+ concentrationconcentration in Fura2/AMFura2/AM -loaded neurons are described. We also describe a method for apoptosisapoptosis detection by immunofluorescenceimmunofluorescence imagingimaging . Finally, a simple method for concurrent measurements of [Ca2+]cyt[Ca2+]cyt and apoptosis in the same neurons is described. |
