Methylation of the 5-cytosine (m(5)C) is a common but not well-understood RNA modification, which can be detected by sequencing of bisulfite-treated transcripts (RNA-BSseq). In this Chapter, we discuss computational RNA-BSseq data analysis methods for transcriptome-wide identification and quantification of m(5)C.
